Top seed Sinner, who claimed his first Grand Slam title at the Australian Open earlier this year, faces Tommy Paul. Meanwhile, 2021 US Open champion Medvedev will take on Portuguese underdog Nuno Borges.
In the women’s draw, French Open champion and world number one Iga Swiatek is aiming to move closer to her second US Open title as she prepares to face Russia’s Liudmila Samsonova.
At 23, Sinner hopes to be the only male player this year to reach the quarter-finals of all four Grand Slams. The Italian previously made it to the last eight in 2022 but was defeated by eventual champion Alcaraz after squandering a match point. With both Djokovic and Alcaraz out of the competition, Medvedev, the fifth seed, remains the only former champion in the draw.
Should Sinner and Medvedev advance, they would meet in the quarter-finals for the fourth time this year. Sinner has already bested Medvedev twice in 2024, including a comeback victory in the Australian Open final, while Medvedev secured a five-set win at Wimbledon.
